We have developed a new bioorthogonal reaction usingTePhe, a tellurophene-based phenylalanine analogue. This reaction, called oxidation-controlled, strain-promoted tellurophene-alkyne cycloaddition (OSTAC), involves mild oxidation of tellurophene to create a reactive species that rapidly cyclizes with bicyclo[6.1.0]nonyne (BCN). OSTAC allows for selective labeling of proteins in complex mixtures and can be combined with other reactions like SPAAC and CuAAC, showing broad potential in biological applications.https://pubs.acs.org/doi/10.1021/jacs.4c07275
