Our new paper presents how N-azidoacetylglucosamine (GlcNAz) can be incorporated into bacterial peptidoglycan for metabolic labeling in Escherichia coli. By the expression of NahK in E. coli, the cytosolic production of UDP-GlcNAz and preferential incorporation into peptidoglycan (PGN) over other sites like poly-β-1,6-N-acetylglucosamine (PNAG), lipopolysaccharide (LPS), or enterobacterial common antigen (ECA) was revealed, providing insights for future development of carbohydrate-based probes and metabolic engineering strategies.https://doi.org/10.1021/acschembio.3c00501
